Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes. After antigen retrieval, incubate samples with the primary antibody for 30 minutes at room temperature.
A suggested positive control is breast carcinoma tissue.

Target Information

GRP94 is an 803 amino acid protein belonging to the heat shock protein 90 family. It functions as a molecular chaperone involved in the processing and transport of secreted proteins. GRP94 and its N-terminal fragment stimulate CTL expansion and maturation of human monocyte-derived dendritic cells (MDDC).
It plays a central role in both innate and acquired immunity, dendritic cell maturation and chemotaxis, antibody production, and cross-priming. GRP94 serves as a potential marker in breast cancer and acts as a peptide acceptor in the endoplasmic reticulum, assisting peptide loading of MHC class I molecules.
Expression of GRP94 suppresses A23187-induced apoptosis and stabilizes calcium homeostasis. GRP94 is expressed in melanoma, liver metastases of colon carcinoma, and human gastric carcinoma BGC-823 cells.
