Target Information
The 26S proteasome is a multicatalytic proteinase complex composed of a 20S core and a 19S regulator. The 20S core consists of four rings with 28 non-identical subunits (two alpha and two beta rings). The 19S regulator includes a base (six ATPase and two non-ATPase subunits) and a lid (up to ten non-ATPase subunits).
Proteasomes are abundant in eukaryotic cells and cleave peptides via an ATP/ubiquitin-dependent, non-lysosomal pathway. The immunoproteasome plays a key role in processing class I MHC peptides.
The PSMC4 gene encodes one of the ATPase subunits of the 19S regulator, belonging to the triple-A ATPase family with chaperone-like activity. It interacts with liver-expressed nuclear receptor family members and gankyrin, a liver oncoprotein. Two transcript variants encoding different isoforms have been identified.
