Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human IL-1alpha. Anti-Human IL-1alpha-specific antibody was purified by affinity chromatography employing an immobilized Human IL-1alpha matrix.

Sandwich ELISA:
To detect Human IL-1alpha by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL is required. In conjunction with PeproTech Biotinylated Anti-Human IL-1alpha (500-P21ABt) as a detection antibody, this allows detection of at least 0.2–0.4 ng/well of Recombinant Human IL-1alpha.

Western Blot:
To detect Human IL-1alpha by Western Blot, use at 0.1–0.2 µg/mL. With compatible secondary reagents, the detection limit for Recombinant Human IL-1alpha is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

Target Information

IL-1 alpha (Interleukin-1 alpha) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. It is encoded by the IL1A gene and signals through IL-1RI and IL-1RII receptors, shared with IL-1 beta. Located on chromosome 2q13, IL-1 alpha plays a crucial role in innate immune defense by triggering production of other proinflammatory cytokines and initiating acute-phase responses.

IL-1 alpha activity is regulated by IL-1 Receptor Antagonist (IL-1RA), which competitively inhibits receptor binding. It is involved in immune responses, inflammation, and hematopoiesis, and is processed and released upon cell injury, inducing apoptosis. The IL1A gene forms part of a cytokine gene cluster and modulates NF-kappaB and MAPK activity, stimulating IL6 and COX-2 expression and nitric oxide production. Elevated IL-1 alpha levels are linked to chronic inflammatory diseases such as rheumatoid arthritis, psoriasis, and multiple sclerosis.
