Product Specific Information

Antibody detects endogenous levels of PAK1 when phosphorylated at Ser199/204 or PAK2 when phosphorylated at Ser192/197.

Target Information

Three isoforms of serine/threonine kinases, designated alphaPAK (p68), betaPAK (p65), and gammaPAK (p62), exhibit a high degree of sequence homology with the S. cerevisiae kinase Ste20, which is involved in pheromone signaling.
The alpha, beta, and gammaPAK isoforms specifically complex with Rac1 and Cdc42 in their active GTP-bound state, inhibiting their intrinsic GTPase activity and leading to autophosphorylation.
There are eight sites of autophosphorylation on gammaPAK, including Ser19, Ser141, and Thr402, and phosphorylation of Ser141 and Thr402 correlates with gammaPAK activation.
Once phosphorylated and their affinity for Rac/Cdc42 reduced, the PAK isoforms disassociate from the complex to seek downstream substrates.
