Product Specific Information

Antibody detects endogenous levels of p53 only when phosphorylated at Thr81.

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or metabolic stress. p53 activation leads to transcription of genes such as p21^waf1^ that regulate G1 or G2/M arrest.
The protein has four main domains: N-terminal transactivation, central DNA-binding, oligomerization, and C-terminal regulatory domains. Phosphorylation at various residues, including Ser15 and Ser392, is crucial for its activation and tumor suppression function.
p53 levels are typically low in normal cells but elevated in transformed or stressed cells, where it contributes to tumor suppression. Mutations in TP53 are common in many human cancers and can abolish its DNA-binding and tumor suppressor activity. Germline TP53 mutations are also associated with cancer-prone syndromes such as Li-Fraumeni syndrome.
