This antibody detects endogenous levels of p53 only when phosphorylated at Thr18.
Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation.
p53 activation leads to transcription of genes such as p21waf1, regulating G1 or G2/M arrest. Structurally, p53 comprises:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerisation domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is crucial for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 also influences aging, oxidative stress, and memory processes.
In normal cells, p53 is expressed at low levels, but is elevated in transformed cell lines, contributing to malignancy. Mutations in TP53 disrupt DNA binding and tumor suppressor activity, and occur in both somatic and germline forms (e.g., Li-Fraumeni syndrome).
