Product Specific Information

The YGIC86 monoclonal antibody reacts with mouse Glycoprotein A Repetitions Predominant (GARP), also known as Lrrc32 or Garpin. GARP is an ~80 kDa glycoprotein expressed on the cell surface. Expression has been reported in placenta, lung, kidney, heart, ovary, liver, skeletal muscle, and pancreas. Protein expression is observed on megakaryocytes, platelets, and activated regulatory T (Treg) cells.
GARP expression on Treg cells is necessary for their suppressive function and may act as a receptor for LAP/TGF beta.

• When analyzing platelets, use Foxp3 Staining Buffer Set (Product #00-5523) for fixation before staining.
• Fixation/permeabilization is not required for Treg cell staining.
• YGIC86 does not cross-react with rat GARP.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on stimulated mouse splenocytes; ≤0.5 µg/test (100 µL final volume).
Cell number can range from 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Light Sensitivity: Protect vial and stained samples from light (photo-induced oxidation sensitive).
Fixation: Samples may be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product #00-8222)
• 1-step Fix/Lyse Solution (Product #00-5333)

Target Information

GARP (Leucine Rich Repeat Containing 32) is a type I membrane protein with 22 leucine-rich repeats and a short intracytoplasmic region. Identified via gene expression profiling of Treg and Th cells after TCR stimulation.
Expressed in CD4+, CD25+, FoxP3+ (Treg) cells, making it a potential cell surface marker for Treg cells.
