Product Specific Information
This 22F6 monoclonal antibody reacts with human, mouse, and canine Helios.
Helios is a zinc-finger transcription factor related to Ikaros, expressed in hematopoietic stem cells and regulatory T cells, but not in mature B cells, dendritic cells, or myeloid cells.
Eight isoforms exist, some with dominant-negative functions associated with lymphoid malignancies such as T-acute lymphoblastic leukemia.
22F6 also cross-reacts with rat Helios.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested using mouse lymph node cells with Foxp3/Transcription Factor Buffer Set (Product #00-5523-00).
Recommended use: 5 µL (0.015 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.
Light Sensitivity: Protect from light to avoid photo-induced oxidation.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54).
Excitation: 488–561 nm
Emission: 775 nm
Laser Compatibility: Blue, Green, Yellow-Green
Filtration: 0.2 µm post-manufacturing filtered
Target Information
Helios belongs to the Ikaros family of zinc-finger transcription factors (Ikaros, Aiolos, Helios).
These proteins regulate lymphocyte development and form homo- or heterodimers with other family members.
Multiple transcript variants exist, though biological validity for some remains unclear.
