Product Specific Information

The MTS510 monoclonal antibody reacts with the mouse Toll-like receptor 4 (TLR4)/MD-2 complex. The Toll family includes type I transmembrane proteins characterized by leucine-rich repeats and a cytoplasmic domain homologous to the IL-1 receptor. TLR2 and TLR4 act as pattern recognition receptors involved in innate immunity and inflammation. TLR4 associates with MD-2 and CD14 to recognize and signal in response to LPS.

In mice, TLR4 is expressed by thioglycolate-elicited peritoneal macrophages, and LPS exposure downregulates surface TLR4/MD-2. The TLR4 gene is defective in C3H/HeJ and C57BL/10ScCr mice, which are LPS hyporesponders.

The MTS510 antibody co-immunoprecipitates MD-2 (~30 kDa) and TLR4 (~100 kDa) and preferentially binds TLR4 associated with MD-2. In contrast, UT41 binds TLR4 regardless of MD-2 association.

Applications Tested: Tested on mouse peritoneal-exudate cells; use ≤2 µg/test (100 µL final volume). Optimize empirically for best performance.

Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54).

Target Information

MD-2 belongs to the Toll/interleukin-1 receptor (TIR) family, which includes Toll-like receptors (TLRs). These receptors recognize pathogen-associated molecular patterns (PAMPs) and link innate and adaptive immunity. TLR4, the main receptor for lipopolysaccharide (LPS), requires MD-2 binding for maximal response. The specificity of this interaction depends on MD-2 species origin; for example, human MD-2 expressed in mouse cells can alter TLR4 response to LPS analogs.
