Product Specific Information
The HTA125 monoclonal antibody reacts with human Toll-like receptor 4 (TLR4). TLR4, a member of the Toll family, is a type I transmembrane protein involved in innate immunity and inflammation. It forms a complex with MD-2 and CD14 for LPS recognition and signaling. TLR4 is expressed on peripheral blood monocytes and shows donor-dependent variability.
Staining Recommendation:
For optimal TLR4 staining, use the lysed whole blood protocol (BestProtocols®) rather than Ficoll-gradient prepared cells, as density gradient separation reduces staining intensity.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated for flow cytometry using normal human peripheral blood cells. Use 5 µL (1 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.
Light Sensitivity: This tandem dye is sensitive to photo-induced oxidation. Protect from light.
Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C with minimal impact on brightness. Clone-specific performance should be empirically determined.
Excitation / Emission / Laser:
488–561 nm excitation; 775 nm emission; compatible with Blue, Green, and Yellow-Green lasers.
Filtration:
0.2 µm post-manufacturing filtered.
Target Information
TLR4 (Toll-like receptor 4) is a key component in pathogen recognition and activation of innate immunity. It is most abundantly expressed in placenta and myelomonocytic leukocytes. TLR4, together with CD14 and LY96, mediates recognition of lipopolysaccharide (LPS) from Gram-negative bacteria, triggering cytokine production for immune response. Mutations in TLR4 affect LPS responsiveness. TLR4 is expressed by monocytes, a subset of B-cells, and placenta.
