Target Information
ROCK1 is a serine/threonine kinase that regulates cell differentiation and migration. The Rho/ROCK pathway is involved in the progression of various human cancers. ROCK1 has been identified as a Caspase-3 substrate.
ROCK, an effector of the small GTPase Rho, contributes to myosin light chain (MLC) activation through two pathways: direct phosphorylation of MLC and phosphorylation/inhibition of MLC phosphatase. This leads to increased cellular contractility necessary for apoptotic membrane blebbing.
ROCK1 consists of an amino-terminal kinase domain and a cysteine/histidine-rich C-terminal inhibitory domain within a pleckstrin-homology region, connected by a variable region containing the Rho-binding domain. During apoptosis, caspase-3 cleaves ROCK1 at a conserved DETD1113/G sequence, removing its inhibitory domain and resulting in constitutive kinase activity. This cleavage induces MLC phosphorylation and membrane blebbing, an early event in apoptosis execution.
