Product Specific Information

The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha).
CD127 complexes with CD132 (common gamma chain) to form the IL-7 receptor (IL-7R), a type I glycoprotein (75–80 kDa) expressed on immature B cells, thymocytes, and most mature T cells. IL-7 binding activates signaling through tyrosine kinase pathways including Jak/STAT, essential for lymphocyte development and T-cell homeostasis.

CD127 expression is down-regulated on CD4+CD25+ regulatory T cells (T regs). These cells, characterized by low CD127 expression, are enriched for regulatory T populations expressing Foxp3 and exhibiting suppressive activity.

Binding of the eBioRDR5 antibody to PBMCs is blocked by recombinant human IL-7. Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry of normal human PBMCs at 5 µL (0.6 µg)/test (100 µL final volume).

Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Product # C001T06F01) is required for optimal staining. Add 5 µL per sample or per 100 µL cell suspension (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information

CD127 (Interleukin-7 receptor, IL-7R) is a glycoprotein regulating lymphopoiesis.
The active receptor is an alpha/gamma chain heterodimer; the gamma(c) chain mediates signal transduction, while the alpha chain determines specific signaling events.
CD127 promotes proliferation of precursor B cells, thymocytes, T cell progenitors, and mature CD4+/CD8+ T cells.
Dysfunction of CD127 is associated with severe combined immunodeficiency.
