Product Specific Information

The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 complexes with CD132 (common gamma chain) to form the IL-7 receptor (IL-7R), a type I glycoprotein expressed on immature B cells, thymocytes, and most mature T cells.

IL-7 binding triggers signal transduction via tyrosine kinase pathways, including Jak/STAT, and plays a crucial role in lymphocyte development and T-cell homeostasis. CD127 expression is down-regulated on CD4+CD25+ regulatory T cells (T regs), and low CD127 expression correlates with Foxp3+ suppressive T reg populations.

Binding of eBioRDR5 to PBMCs is blocked by pre-incubation with recombinant human IL-7 (Product #14-1079-80).

Applications Tested

Pre-titrated and validated for flow cytometry on normal human peripheral blood cells.
Use at 5 µL (0.6 µg)/test, defined as the antibody amount for staining in 100 µL final volume.
Cell number: 10⁵–10⁸ cells/test.

Important Notes

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Recommended: Invitrogen LIVE/DEAD Fixable Dead Cell stains.
• Use CellBlox Plus Blocking Buffer for optimal staining (Product #C001T06F01).
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD127 (Interleukin-7, IL-7) is a glycoprotein regulating lymphopoiesis. The functional receptor is an IL7R alpha/gamma heterodimer.

• Gamma chain (γc): Activates signal transduction
• Alpha chain: Determines specific signaling events

CD127 promotes proliferation of precursor B cells, thymocytes, T cell progenitors, and mature CD4+/CD8+ T cells.
Defects in CD127 function are associated with severe combined immunodeficiency (SCID).
