Product Specific Information

The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 forms a complex with CD132 (common gamma chain) to generate the IL-7 receptor (IL-7R). It is a type I glycoprotein (75–80 kDa) expressed on immature B cells, thymocytes, and most mature T cells.

Binding of IL-7 activates tyrosine kinase pathways including Jak/STAT, essential for lymphocyte development and T-cell homeostasis. CD127 expression is down-regulated on CD4+CD25+ regulatory T cells (T regs), making it a useful marker for identifying true T reg populations enriched for Foxp3 expression and suppressive activity.

Binding of eBioRDR5 to PBMCs is blocked by recombinant human IL-7 (Product #14-1079-80).

Each product includes:

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry at 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Product #C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD127 (Interleukin-7 Receptor alpha, IL7R) regulates lymphopoiesis through its alpha/gamma chain heterodimer receptor complex. The gamma(c) chain activates signal transduction, while the alpha chain mediates specific signaling events. CD127 promotes proliferation of precursor B cells, thymocytes, and mature T cells.

Dysfunction of CD127 is associated with severe combined immunodeficiency (SCID) and T cell negative/B cell negative/NK positive SCID.
