Product Specific Information
Store vial at -20°C prior to opening. Aliquot contents and freeze at -20°C or below for extended storage. Avoid freeze–thaw cycles. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4°C as an undiluted liquid. Dilute only prior to immediate use.
Anti-IL-22 is an IgG fraction antibody purified from monospecific antiserum by a multi-step process including delipidation, salt fractionation, and ion-exchange chromatography, followed by extensive dialysis against the specified buffer. This antibody is specific for human IL-22 protein. A BLAST analysis suggests cross-reactivity with IL-22 from human sources based on 100% homology with the immunizing sequence. Cross-reactivity with IL-22 from other species has not been determined.
Target Information
IL-22, also known as IL-10-related T-cell derived inducible factor, is an alpha-helical cytokine belonging to the IFN-IL-10 family, which includes IL-19, IL-20, IL-24, IL-26, IL-28, IL-29, and type I and II interferons. IL-22 is mainly produced by activated T cells and NK cells. In humans, the IL-22 gene is located on chromosome 12q and is structurally related to IL-10.
IL-22 acts through a heterodimeric receptor complex consisting of IL-22R1 and IL-10R2. IL-22R1 also binds IL-20 and IL-24, while IL-10R2 binds IL-10, IL-27, IL-28, and IL-29. Binding of IL-22 to its receptor induces JAK-STAT signaling. A soluble receptor, IL-22BP, can antagonize IL-22 binding and signaling.
IL-22 primarily targets nonimmune cells such as keratinocytes, hepatocytes, and epithelial cells of the skin, digestive, and respiratory systems. It functions as an effector cytokine of the Th17 lineage and, along with IL-17A and IL-17F, regulates genes involved in innate skin immunity. IL-22 promotes wound healing, induces antimicrobial proteins, and is critical for defense against extracellular pathogens. It is upregulated during chronic inflammation and activates intracellular kinases and transcription factors, including STAT3.
