Product Specific Information

The monoclonal antibody P1H12 recognizes CD146, also known as MUC18, s-endo, Endo-CAM, and Mel-CAM, a member of the Ig superfamily.
CD146 is expressed on endothelial cells, bone marrow fibroblasts, some tumors (especially melanoma), mesenchymal stromal cells, and endometrial stromal cells.
Expression on circulating blood cells is limited to a subset of T cells rather than endothelial cells. The protein is heavily glycosylated, with over 50% of its mass derived from carbohydrates.

Target Information

CD146 (MCAM) is an integral membrane glycoprotein of the immunoglobulin superfamily.
It is expressed at cell-cell junctions in endothelial cells and various normal and malignant cell types.
CD146 functions as a calcium-independent adhesion molecule and signal transduction mediator, recruiting Fyn kinase and promoting tyrosine-phosphorylation of proteins involved in actin cytoskeleton assembly.
It plays a key role in endothelial cohesion, melanoma metastasis, and neural crest cell adhesion during embryonic development.
CD146 also acts as a surface receptor that triggers phosphorylation of FYN and PTK2, and transiently increases intracellular calcium concentration.
