Product Specific Information

This NJTEN3 monoclonal antibody reacts with the pro-form of mouse IL-1 beta, a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. It recognizes only the pro-form and not the active (cleaved) form.
Pro-IL-1 beta (31 kDa) is synthesized in response to inflammatory stimuli and requires inflammasome activation for cleavage into the active 17 kDa form. IL-1 beta is secreted via a non-classical pathway and signals through IL-1RI, shared with IL-1 alpha. These cytokines play key roles in innate immunity and inflammation regulation.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested on stimulated mouse peritoneal macrophages using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00) and BestProtocols® Protocol A.
Use ≤0.5 µg/test (100 µL final volume). Adjust cell number empirically (10⁵–10⁸ cells/test). Titrate antibody for optimal performance.

Target Information

Interleukin-1 beta (IL-1β) is a proinflammatory cytokine produced by monocytes, macrophages, and dendritic cells. It is synthesized as a 31 kDa pro-form and processed to an active 17 kDa form by caspase-1 after inflammasome activation. IL-1β signals through IL-1RI and IL-1RII (shared with IL-1α), promoting cytokine production and acute-phase responses. IL-1RA can inhibit IL-1β activity through receptor competition. Elevated IL-1β is linked to chronic inflammatory diseases, and IL-1β neutralization has therapeutic potential.
