Product Specific Information

The MR1 monoclonal antibody reacts with mouse CD154, a 39 kDa transmembrane glycoprotein also known as CD40 ligand (CD40L). CD154 is transiently expressed by activated T cells. Through binding to CD40 on antigen-presenting cells (B cells, monocytes/macrophages, dendritic cells), it plays a key role in T cell–APC interactions, B cell activation, and cell cycle entry.

For flow cytometry staining, enriched T cells or CD4 cells should be stimulated prior to staining with MR1.
This antibody is validated for flow cytometric analysis of stimulated mouse splenocytes and can be used at ≤0.5 µg/test (100 µL final volume). Cell number may vary from 10⁵–10⁸ cells/test. Optimization by titration is recommended.

Fluorophore Information:
eFluor® 450 is an alternative to Pacific Blue®, excited by the violet laser (405 nm) and emitting at 445 nm. Ensure the cytometer is compatible with this fluorochrome.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD40 Ligand (CD40-L or CD154) is a membrane glycoprotein expressed on T cells. It stimulates B cell proliferation and immunoglobulin secretion in the presence of cytokines, induces cytokine production and tumoricidal activity in monocytes, and co-stimulates activated T cells to produce IFN-γ, TNF-α, and IL-2.
