The monoclonal antibody H194-112 recognizes mouse CD26, a thymocyte-activating molecule (THAM).
CD26 is a surface-expressed and soluble ectoenzyme (dipeptidyl peptidase IV) that cleaves dipeptides from the amino terminus of proteins, including CXCL12, CCL22, neuropeptide Y, and glucagon.

CD26 is involved in:

• Adhesion
• Apoptosis
• Disease regulation
• Engraftment

Expression is regulated during thymic development at the mRNA level. CD26 protein is found on thymocytes, a subset of resting and activated B and T cells, NK cells, and epithelial cell types.

On human cells, CD26 is more abundant on Th2 cells. Functional CD26 is expressed on Sca-1⁺c-Kit⁺lin⁻ cells, correlating with progenitor cell mobilization. Inhibition of CD26 enhances allogenic stem cell homing and engraftment.

This antibody has activating function and is validated for flow cytometric analysis of mouse splenocytes or thymocytes.
Recommended usage: ≤1 µg per test (100 µL total volume, 10⁵–10⁸ cells/test).
Careful titration is advised for optimal performance.

CD26 (Dipeptidyl Peptidase IV, DPP IV) is a homodimeric atypical serine protease belonging to the prolyl oligopeptidase family.
It is expressed on lymphocytes and upregulated during T-cell activation, also found on activated B cells, NK cells, and epithelial tissues.

CD26 functions include:

• T-cell activation
• Cell adhesion (fibronectin, collagens)
• HIV infection mediation

CD26 corresponds to adenosine deaminase complexing protein-2 and the T-cell activation antigen CD26.
It is a membrane glycoprotein and serine exopeptidase that cleaves X-proline dipeptides from the N-terminus of polypeptides.

Alterations in CD26 activity are linked to malignant transformation, with increased enzymatic activity correlating with tumor grade and severity.
Changes in CD26 density have been observed in monocytes and CD4⁺ T cells during rheumatoid arthritis and systemic lupus erythematosus.
