  • Domain: The RING finger domain functions as a structural scaffold, bringing two clusters of positively charged residues close together to mimic a bipartite nuclear localization signal (NLS).
  • Function: E3 ubiquitin-protein ligase mediating ubiquitination and proteasomal degradation of target proteins such as JUN and p53 (TP53). It abolishes p53-dependent transcription and apoptosis by ubiquitinating p53 independently of MDM2 or RCHY1.
  • Pathway: Protein modification; protein ubiquitination.
  • Similarity: Belongs to the COP1 family; contains 1 RING-type zinc finger and 7 WD repeats.
  • Subunit: Forms homodimers via coiled coil domain. Component of DCX DET1-COP1 ubiquitin ligase complex (RBX1, DET1, DDB1, CUL4A, COP1). Isoform interactions vary with CUL proteins. Interacts with JUN, JUNB, JUND, and p53 (TP53).
  • Tissue Specificity: Ubiquitously expressed at low level; higher expression in testis, placenta, skeletal muscle, and heart.
  • Induction: By p53/TP53.
