Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. p53 upregulation activates transcription of genes like p21waf1, leading to G1 or G2/M arrest.
Structurally, p53 includes:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerisation domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is key for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays a crucial role in preventing proliferation of damaged cells, acting as an anticancer agent. It is expressed at low levels in normal cells but highly expressed in transformed cell lines, contributing to malignancy. Mutations in TP53 result in loss of tumor suppressor activity and are implicated in various cancers, including germline mutations in Li-Fraumeni syndrome.
