Target Information
The human AF9 gene (MLLT3) is one of the most common fusion partner genes with the ALL1 gene (MLL) at 11q23, resulting in the t(9;11)(p22;q23) translocation.
AF9 spans over 100 kb and contains two breakpoint cluster regions (BCRs):
- BCR1: within intron 4 (site A)
- BCR2: spans introns 7 and 8 (site B)
Structural elements identified in AF9 include:
- DNA topo II cleavage sites
- DNase I hypersensitive sites in BCR2
- Scaffold associated regions (SARs) centromeric to cleavage sites (SAR1 in intron 4, SAR2 in exons 5–7)
These regions share structural features with the MLL breakpoint cluster region.
A DNA breakage and repair model for nonhomologous recombination between MLL and AF9 has been proposed.
