Product Specific Information

The eBio1A6 monoclonal antibody reacts with human thymic stromal-derived lymphopoietin receptor (TSLPR), a ~50 kDa protein similar to the common gamma-chain. TSLPR forms a high-affinity receptor complex with IL-7Rα (CD127) to bind TSLP. It is expressed by monocytes and CD11c⁺ dendritic cells, where TSLP binding induces Th2-attracting chemokines CCL17 and CCL22.

Activation of dendritic cells via TSLPR increases secretion of IL-4, IL-5, and IL-13, contributing to CD4⁺ T cell homeostasis. In mice, TSLPR deficiency alone does not affect lymphocyte numbers, but combined deficiency with the common gamma-chain reduces lymphocyte counts. The eBio1A6 antibody can cross-block another anti-human TSLPR antibody, 1D8.

Applications Reported: Flow cytometry and Western blotting (WB).
Applications Tested: Validated in flow cytometric analysis using human TSLPR-transfected Ba/F3 cells. Recommended ≤0.5 µg/test in 100 µL volume, with 10⁵–10⁸ cells/test. Optimize titration for best performance.
Purity: >90% (SDS-PAGE).
Aggregation: <10% (HPLC).
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Thymic stromal lymphopoietin (TSLP) drives dendritic cell maturation and activation, supports regulatory T cell selection, and maintains peripheral CD4⁺ T cell homeostasis. It also promotes B cell progenitor growth and differentiation to IgM⁺ stage. TSLP receptor (TSLP-R) binds TSLP with low affinity; co-expression with IL-7Rα forms a high-affinity, functional heteromeric complex that signals via STAT5 and is inhibited by SOCS-1.
