Product Specific Information

The monoclonal antibody Moushi101 recognizes mouse CD101 (also known as IGSF2, V7), a transmembrane glycoprotein with seven Ig-like domains and a short cytoplasmic tail containing phosphorylation sites. Cross-linking CD101 can antagonize TCR/CD3-mediated lymphocyte activation.

Expression:
CD101 is expressed on monocytes, granulocytes, dendritic cells, Langerhans cells, and activated T cells. It is also found on a subset of CD25⁺Foxp3⁺ regulatory T cells with high suppressive activity.

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Tested on mouse bone marrow cells. Recommended at ≤ 0.25 µg per test (100 µL final volume). Cell number should be determined empirically (10⁵–10⁸ cells/test). Titrate for optimal performance.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in:

• IC Fixation Buffer (Product #00-8222-49), or
• 1-step Fix/Lyse Solution (Product #00-5333-54)
  for up to 3 days at 4°C in the dark with minimal loss of brightness and FRET efficiency.

Target Information

CD101 (V7 antigen) is a member of the immunoglobulin superfamily and exists as a disulfide-linked homodimer. It is expressed on monocytes, granulocytes, and dendritic cells (Langerhans-like cells expressing HLA-DR, CD1a, CD1c). Expression on T lymphocytes contributes to cell activation. CD101⁺CD28⁺ cells respond strongly to CD28 signaling, and co-stimulation with anti-CD28 or suboptimal anti-CD3 enhances proliferation.

Mouse CD101 is expressed on a subset of CD4⁺CD25⁺Foxp3⁺ regulatory T cells with strong suppressive activity. In human PBMCs, approximately 30% of Foxp3⁺ cells express CD101, though enhanced suppressive activity has not been confirmed in humans.
