Product Specific Information
Reconstitution:
Centrifuge the vial briefly before opening. Reconstitute the lyophilized protein in sterile, distilled water to 0.1–1.0 mg/mL. Apportion into working aliquots and store at -20°C. Further dilutions should be made in low endotoxin medium or buffered solution containing carrier protein (e.g., heat-inactivated FCS or tissue culture grade BSA).Storage:
Store lyophilized protein at 2–8°C, preferably desiccated. Upon reconstitution, divide into aliquots and store at -20°C. Avoid repeated freeze-thaw cycles.
Target Information
TNF-alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. Secreted mainly by macrophages, it binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR receptors. TNF-alpha regulates immune cell activity, proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It forms multimers (dimers, trimers, or pentamers) but appears as a single 17 kDa band under non-reducing SDS-PAGE conditions. Knockout studies suggest neuroprotective roles and tumor necrosis induction in mice. TNF-alpha also mediates immune responses to bacterial, viral, parasitic, and fungal infections, and contributes to tumor cell cytolysis—synergistic with interferon-gamma.
It has been implicated in autoimmune diseases, insulin resistance, and cancer.
