Applications and Tested Dilution

• Flow Cytometry (Flow): Tested at ≤0.2 µg/test
  A test is defined as the amount of antibody that stains a cell sample in a final volume of 100 µL (10⁵–10⁸ cells/test).
  Antibody should be titrated for optimal performance.

Product Specific Information

The 53-7.3 monoclonal antibody reacts with mouse CD5, a 67 kDa protein expressed by most thymocytes, mature T cells, and a subset of B cells (B-1 cells). CD5 functions as a counter-receptor for CD72 and plays a role in T–B cell interactions.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis of mouse splenocytes.
Applications Tested: Suitable for ≤0.2 µg/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample (regardless of number of NovaFluors).
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology based on nucleic acid dye structures engineered for minimal spillover and spread.

Excitation: 552 nm
Emission: 690 nm
Laser: 561 nm (Yellow)

Internal testing shows strain-dependent staining:

• Non-specific staining observed in SJL mice
• Not observed in BALB/c or C57BL/6 mice

Target Information

CD5 is a 67 kDa transmembrane glycoprotein expressed on mature T-lymphocytes, thymocytes, and subsets of activated B cells. It regulates T-cell proliferation and is associated with thymus cancer and Richter’s Syndrome.
