Product Specific Information

The 53-7.3 monoclonal antibody reacts with mouse CD5, a 67 kDa protein expressed by most thymocytes, mature T cells, and a subset of B cells. CD5 serves as a counter-receptor for CD72 and participates in T-B cell interactions.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications

• Reported for use in flow cytometric analysis
• Tested on mouse splenocytes at ≤0.25 µg/test (100 µL final volume)

Note:

• Determine cell number empirically (10⁵–10⁸ cells/test).
• Titrate antibody for optimal performance.
• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01)

• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample, regardless of the number of NovaFluors
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells)

NovaFluor conjugates are based on Phiton™ technology utilizing novel nucleic acid dye structures engineered for optimized fluorescent signatures with reduced spillover and spread.

Excitation: 552 nm

Laser: 561 nm (Yellow)

Note:
NovaFluor Yellow 660 shows non-specific staining of B cells in SJL mice, but not in BALB/c or C57BL/6 strains.

Target Information

CD5 is a 67 kDa single-chain transmembrane glycoprotein found on mature T-lymphocytes, thymocytes, and subsets of activated B cells. It regulates T-cell proliferation and is associated with thymus cancer and Richter’s Syndrome.
