Target Information
LCK는 Src-family tyrosine kinase에 속하며, T-cell receptor (TCR) 신호 전달에 필수적인 역할을 합니다. TCR 자극 시 LCK는 zeta 서브유닛의 ITAM 모티프를 인산화하여 ZAP70의 SH2 도메인 결합 부위를 형성합니다. ZAP70은 LCK에 의해 인산화 및 활성화되어 LAT 어댑터 단백질을 통한 신호 전달 플랫폼을 생성합니다. 대부분의 LCK는 세포막에 존재하지만, 일부는 골지체에 위치하여 약한 TCR 자극 시 Raf 활성화에 기여할 수 있습니다. LCK는 N-말단의 myristylation 및 palmitylation 부위, PTK 도메인, SH2 및 SH3 도메인을 포함하며, 이는 인산화티로신 및 프롤린 풍부 모티프와의 단백질 상호작용을 매개합니다. 또한 LCK는 다양한 자극에 의해 유도되는 세포사멸 조절에 관여하지만, death receptor에 의한 세포사멸에는 관여하지 않습니다. LCK 관련 질환으로는 Immunodeficiency 22 및 CD45 deficiency가 있으며, 다양한 isoform을 암호화하는 대체 스플라이싱 변이가 보고되어 있습니다.
