Product Specific Information

Description:
The eBio124-1D1 monoclonal antibody reacts with human CD7 (gp40, Leu9), a 40 kDa receptor and member of the immunoglobulin gene superfamily. The N-terminal domain (aa1–107) is homologous to Ig kappa light chains, while the C-terminal region is proline-rich and forms a stalk projecting the Ig domain.

CD7 is expressed on most immature and mature T lymphocytes, T cell leukemias, NK cells, and a subset of B cells. Cross-linking of CD7 modulates T cell and NK cell activity, influencing calcium flux, adhesion molecule expression, cytokine secretion, and proliferation. CD7 associates with phosphoinositol 3′-kinase, inducing D-3 phosphoinositide production and tyrosine phosphorylation.

A subpopulation of CD34(+) CD38(-) cord blood cells expressing high CD7 levels shows lymphoid differentiation potential, generating B cells, NK cells, and dendritic cells but not myeloid or erythroid cells. CD7 thus helps distinguish primitive lymphoid progenitors from pluripotent stem cells.

CD7 may also cooperate with CD28 in Treg function, as double-deficient mice (CD28⁻/⁻ CD7⁻/⁻) show reduced Treg numbers and suppressive activity.

Applications Tested: Pre-titrated for use at 5 µL (0.8 µg) per 100 µL test sample (10⁵–10⁸ cells/test).

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
    • Add 5 µL per sample before combining with cells.
    • For single-color controls: 5 µL per 100 µL cell sample.
