Product Specific Information
The GL1 monoclonal antibody reacts with mouse CD86 (B7-2), an ~80 kDa surface receptor and member of the B7 costimulatory family. CD86 is expressed at low levels on B cells, macrophages, and dendritic cells, and is upregulated through stimuli such as BCR complex, CD40, and cytokine receptors. It is also expressed by activated mouse T cells and peritoneal cells.
CD86 interacts with T-cell surface molecules CD28 and CTLA-4 (CD152), playing a critical role in T-B cell crosstalk, T-cell costimulation, autoantibody production, and Th2-mediated Ig production. Upregulation kinetics indicate CD86’s major contribution during the primary immune response phase.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometry on stimulated mouse splenocytes. Recommended ≤1 µg/test (100 µL final volume). Optimal titration required.
Fluorochrome Information:
Super Bright 436 is excited by a 405 nm violet laser and emits at 436 nm. Recommended detection with a 450/50 bandpass filter or equivalent. Ensure instrument compatibility.
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information
CD86, along with CD80, serves as a ligand for CTLA4 and CD28, acting as a costimulatory molecule in T-cell activation during antigen presentation. Insufficient co-stimulation via CD86/CD80 can induce tolerance. CD86 is a type I membrane protein of the immunoglobulin superfamily, expressed by antigen-presenting cells.
Binding of CD86 with CD28 provides a costimulatory signal for T-cell activation, whereas binding with CTLA-4 inhibits T-cell activation, reducing immune response. Alternative splicing produces multiple isoforms. CD86 dysfunction is associated with diseases such as gallbladder squamous cell carcinoma and myocarditis.
