Product Specific Information

Directed against human SMAD4 protein.
A BLAST analysis indicates cross-reactivity with SMAD4 from human, mouse, and Xenopus based on 100% homology with the immunizing sequence.
Reactivity against homologues from other species is not known.
Target Information
Smad4 is encoded by the Smad4 gene located on chromosome 18 in humans. Also known as Smad family member 4, Sma- and Mad-related protein 4, or Mothers against decapentaplegic (MAD) homolog 4.
Smad4 contains N-terminal MH1 and C-terminal MH2 domains involved in DNA binding and protein interactions.
Binding of TGF-beta and BMP ligands to their receptors leads to phosphorylation of R-Smads (1, 2, 3, 5, and 8), which then form heterotrimers with Smad4 and translocate to the nucleus.
Smad4 functions as a tumor suppressor by transcriptionally regulating genes such as Cyclin D1 (downregulated) and collagen (upregulated), influencing cell proliferation.
Dephosphorylation controls nuclear export and nucleocytoplasmic dynamics of Smads.
