Description:
The MIH18 monoclonal antibody reacts with human B7-DC, also known as PD-L2. B7-DC is a member of the B7 family with a predicted molecular weight of approximately 25 kDa and belongs to the immunoglobulin superfamily. It is primarily expressed by dendritic cells and monocytes/macrophages. Unlike other B7 family members, B7-DC binds PD-1 rather than CD28/CTLA-4. The PD-1 and B7-DC/PD-L2 interaction is involved in T cell costimulation or suppression depending on activation state. MIH18 is a blocking antibody useful for studying PD-L2 function and expression in human immune cells.

Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of mature dendritic cells and peripheral blood cells. Recommended use: 5 µL (0.125 µg) per test. A test is defined as the amount of antibody required to stain a cell sample in 100 µL final volume. Cell number can vary from 10⁵–10⁸ cells/test.

Target Information

Programmed death-ligand 2 (PD-L2, B7-DC) is a B7 ligand family member that, along with PD-L1, binds PD-1. It is mainly expressed in dendritic cells but can be induced in other immune and non-immune cells. PD-L2 expression is upregulated by IL-4 and to a lesser extent by TNF-α and IFN-γ. It shows 2–6 times higher affinity for PD-1 than PD-L1. PD-1 and its ligands act as inhibitory immune checkpoint molecules, regulating T cell activation and cytokine secretion. PD-L2 may also stimulate T cell proliferation independently of PD-1, suggesting additional receptors such as RGMb. The PD-L2–RGMb interaction plays a role in respiratory tolerance via BMP and neogenin signaling. The human PD-L2 monomer includes a 201 aa extracellular domain, 21 aa transmembrane domain, and 32 aa cytoplasmic domain.
