Product Specific Information
The FN-3 monoclonal antibody recognizes human fibronectin, a high molecular weight extracellular matrix protein (~400 kDa) existing in extracellular and soluble plasma forms. It functions in cell adhesion and migration via integrins and associates with collagen, actin, and fibrins. Fibronectin levels can vary in malignancies; for example, expression decreases in lung carcinomas. FN-3 antibody recognizes a determinant on human cellular but not plasma fibronectin, unaffected by trypsin treatment, and cross-reacts with bovine fibronectin.
Applications Reported: Immunohistochemical staining of formalin-fixed paraffin-embedded (FFPE) tissue sections and immunoblotting.
Applications Tested: Tested by IHC on FFPE human placenta using IHC Antigen Retrieval Solution - Low pH (Product # 00-4955-58), usable at ≤10 µg/mL. Recommended to titrate for optimal assay performance.

Target Information
Fibronectin is a disulfide-bonded dimer (230–250 kDa) found in connective tissues and vessels as an insoluble, cross-linked protein forming high molecular mass polymers. It is abundant during embryonic development and tissue remodeling and also exists as a soluble plasma protein. Fibronectin domains interact with collagen, heparin, and integrins, facilitating cell adhesion and migration in processes such as embryogenesis, wound healing, coagulation, host defense, and metastasis.
In lung cancer, fibronectin expression is increased, particularly in non-small cell lung carcinoma. Its interaction with carcinoma cells enhances tumorigenicity and resistance to apoptosis induced by chemotherapy. The fibronectin gene undergoes alternative splicing in three regions, potentially generating up to 20 transcript variants.
