The monoclonal antibody 8B6 recognizes human Placental Alkaline Phosphatase (PLAP), a 130 kDa enzyme expressed by placental syncytiotrophoblastoid cells. This enzyme exhibits slightly different kinetics from other phosphatases and is found both in the plasma membrane and cytoplasm.
Expression: Found in germ line cancers (ovarian, testicular), human embryonic stem cells, and bone marrow-derived mesenchymal stromal cells.
Isozymes: Reacts with both Regan and Nagao types.

Target Information
Placental Alkaline Phosphatase (PLAP) plays a role in regulating inflammatory processes. Four related alkaline phosphatases exist: intestinal, placental, placental-like, and liver/bone/kidney types. PLAP (Regan and Nagao isoenzymes) is expressed in the placenta during the third trimester and serves as a marker for testicular germ cell tumors.
PLAP-positive somatic cell tumors also express epithelial membrane antigen (EMA). Although proposed to be involved in matrix mineralization, PLAP-deficient mice show normal skeletal development. PLAP has been linked to hypophosphatasia, a disorder involving hypercalcemia and skeletal defects, with severity depending on specific mutations.
