Description:
The monoclonal antibody AC22 recognizes EZH2 (Enhancer of zeste homolog 2), a member of the polycomb group (PcG) proteins responsible for repression of gene expression through recognition and modification of histone methylation and chromatin structure. EZH2 is the catalytic subunit of the polycomb repressive complex 2 (PRC2), directly repressing gene expression through methylation of histone H3 on lysine 27 (H3K27) as well as direct protein targets such as GATA4.
PRC2-mediated gene repression is essential for proper development and differentiation. Underexpression of EZH2 is linked to abnormal developmental patterning and loss of stem cell pluripotency, while overexpression correlates with poor prognosis in melanoma, breast, and prostate cancers.

This AC22 antibody has been reported for use in immunoprecipitation, western blotting, and immunocytochemistry.
Applications Tested:
Tested by immunocytochemistry of methanol-fixed cells, effective at ≤5 µg/mL. Recommended to titrate for optimal assay performance.
Target Information
EZH2 (Histone-lysine N-methyltransferase) is a member of the Polycomb-group (PcG) family. PcG proteins form multimeric complexes maintaining transcriptional repression across cell generations. EZH2 interacts with embryonic ectoderm development protein, VAV1 oncoprotein, and X-linked nuclear protein. It may play roles in hematopoietic and central nervous systems. Mutations in EZH2 can cause Weaver syndrome (WVS).
