Product Specific Information

The monoclonal antibody 12H6 recognizes human P-cadherin (placental-cadherin), a calcium-dependent cell-cell adhesion protein. This 118 kDa transmembrane protein was originally named for its expression in mouse placental tissue, though it is not expressed in human placental tissue.

The function of P-cadherin depends on its association with cytoplasmic catenins (α, β, γ), which determine the strength and signaling of cell-cell adhesion. Expression is more restricted than E-cadherin, being localized to the basal proliferative cell layer of stratified epithelia. During development, expression occurs in proliferating tissues such as hair follicle keratinocytes and ductal mammary tissue.

P-cadherin can be secreted from epithelial cells during late pregnancy stages. Its expression correlates with poor prognosis in breast cancer, while reduced expression in melanomas and squamous cell carcinomas may also indicate poor prognosis.

Target Information

P-cadherin (Cadherin-3) is a Type 1 cadherin protein belonging to the cadherin superfamily. These are single-pass transmembrane glycoproteins with five extracellular cadherin repeats and an intracellular domain binding p120-catenin and β-catenin.

Cadherins mediate calcium-dependent cell-cell adhesion and play roles in development, wound healing, cell signaling, growth, and differentiation.

P-cadherin is expressed in human fetal tissues and adult basal epithelial cells, including epidermis, breast, prostate, ovary, cervix, hair follicle, and corneal endothelium. It is also found in embryonic stem cells and mammary gland stem cells.

Overexpression is linked to poor prognosis in several carcinomas (breast, prostate, ovary, colon, stomach). Mutations in CDH3 cause congenital hypotrichosis with juvenile macular dystrophy.
