Product Specific Information

This antibody reacts to Histone H2AX protein, independent of post-translational modifications, and shows no cross-reactivity with other histone proteins.
Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. These systems clone specific antibody DNA sequences from immunoreactive rabbits, followed by screening to select optimal clones for production.

Target Information

Histone H2A.X (H2AX) is a member of the histone H2A family, one of four core histones forming the nucleosome core particle.
In eukaryotes, DNA double-strand breaks (DSBs) trigger phosphorylation of serine 139 at the C-terminus of H2AX, resulting in γ-H2AX. This phosphorylation can be detected by Western blotting or immunofluorescence, indicating DSB frequency.
Phosphorylation is mediated by phosphatidylinositol 3-kinases, including ATM, DNA-PK, and ATR.
H2A.X consists of 143 amino acids and is synthesized in both G1 and S phases. Its mRNA occurs in two forms: a 1600-base polyA-containing form and a 575-base form lacking polyA.
Histone H2A.X maps to chromosome 11q23.2–q23.3 and contributes to DNA structure formation. The variant H2A.X regulates MDC1 interaction at DNA damage sites.
