Immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
BCL-2 is a key regulator of apoptosis that can inhibit or promote cell death. Members of the BCL-2 family dimerize to modulate apoptosis. For example, Bag1 forms a heterodimer with BCL-2, enhancing its anti-apoptotic effect. Bax and Bak promote cytochrome c release from mitochondria, initiating apoptosis. Overexpression or constitutive expression of BCL-2, such as through translocation to the Ig heavy chain locus, is associated with follicular lymphoma. High BCL-2 alpha expression is observed in neoplastic germinal centers of follicular lymphomas, while normal centers are negative. Two transcript variants of BCL-2 differ in their C-terminal ends. Overexpression has been linked to cancers including B-cell lymphoma and prostate cancer.
