Target Information

Dihydropyridine receptor (DHPR) is a surface membrane protein essential for excitation-contraction coupling in striated muscle. DHPR and the sarcoplasmic reticulum ryanodine receptor (RyR) form key intracellular junctions where surface membrane depolarization triggers Ca²⁺ release from internal stores. The DHPR alpha1-subunit contains a cytoplasmic loop involved in RyR interaction, and its phosphorylation is thought to influence DHPR-RyR functional coupling. Mutations in DHPR alpha1 can cause excitation-contraction uncoupling, leading to muscular dysgenesis and reduced Ca²⁺ current density in RyR-deficient cells.
