Product Specific Information
The eBioHMb1-1 monoclonal antibody reacts with mouse and rat CD29 (integrin beta 1), a 110–120 kDa member of the beta integrin family expressed by leukocytes, endothelial, smooth muscle, and epithelial cells.
CD29 forms non-covalent complexes with alpha integrins (CD49a–f) to generate VLA-1 through VLA-6, and also interacts with CD51. These heterodimers mediate adhesion, trafficking, proliferation, and differentiation.
Integrins containing CD29 bind to extracellular matrix proteins (collagen, laminin, fibronectin, vitronectin) and can interact with cellular receptors such as VCAM-1 and MadCAM-1.
Each product contains:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse bone marrow cells at ≤0.2 µg/test (100 µL final volume).
Cell number range: 10⁵–10⁸ cells/test. Antibody should be titrated for optimal performance.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
- Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample before combining with cells.
- For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).
NovaFluor Technology:
Phiton™ technology enables engineered fluorescent signatures minimizing spillover and spread effects.
Excitation: 552 nm; Emission: 700 nm; Laser: 561 nm (Yellow).
Strain-specific Observation:
NovaFluor Yellow 700 shows non-specific B cell staining in SJL mice; not observed in BALB/c or C57BL/6 strains.
Target Information
ITGB1 (Integrin Subunit Beta 1, CD29):
A 110 kDa cell surface glycoprotein expressed in various cells including leukocytes. Forms heterodimers with alpha chains (α1–α6) determining binding properties of VLA integrins.
Functions as a cell adhesion molecule, platelet component, and extracellular matrix receptor.
Decreased ITGB1 expression correlates with multidrug resistance in tumors.
Up-regulated during leukocyte migration and involved in immune cell trafficking and inflammatory responses.
Two main isoforms: β1A (ubiquitous except muscle) and β1D (cardiac/skeletal muscle).
