Product Specific Information

The 1B1 monoclonal antibody reacts with mouse CD1d, a 48 kDa glycoprotein structurally similar to MHC class I molecules. CD1d associates with beta2-microglobulin and functions analogously to MHC Class II. 1B1 detects CD1d at varying levels on mouse leukocytes.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Tested on mouse splenocytes at ≤ 0.8 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
  Optimal performance requires titration in the assay of interest.

Each product contains:

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) provides improved formulation and is required for optimal staining with NovaFluor conjugates.

• Add 5 µL per sample before combining antibody cocktails with cells.
• For single-color controls: use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.

Internal testing shows non-specific staining of B cells in SJL mice, not observed in BALB/c or C57BL/6 strains.

Target Information

CD1d is a heavy chain associated with beta-2 microglobulin on cortical thymocytes. It is the sole group 2 member of the CD1 family of MHC-like glycoproteins.
CD1 proteins present lipid and glycolipid antigens to T cells and differ by cellular localization and ligand specificity.
The CD1d gene product localizes to late endosomes and lysosomes via a tyrosine-based motif.
Diseases associated with CD1D include Mycobacterium malmoense infection and autoimmune disorders.
