Product Specific Information

The 1B1 monoclonal antibody reacts with mouse CD1d, a 48 kDa glycoprotein structurally homologous to MHC class I molecules. CD1d associates with beta2m and functions similarly to MHC Class II. 1B1 detects CD1d at varying levels on mouse leukocytes and recognizes beta2m-associated CD1d.

  • Applications Tested: Tested on mouse splenocytes at ≤0.6 µg/test (100 µL final volume, 10⁵–10⁸ cells/test)
  • Excitation: 488–561 nm
  • Emission: 578 nm

  • Filtration: 0.2 µm post-manufacturing filtered

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.

  • Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology, utilizing novel nucleic acid dye structures for engineered fluorescent signatures with minimized spillover and spread.

Target Information

CD1d is a heavy chain associated with β2-microglobulin on cortical thymocytes. It can also be expressed independently on human intestinal epithelial cells. CD1d is the sole group 2 member of the CD1 family of MHC-like glycoproteins. These proteins present lipid and glycolipid antigens to T cells. The CD1 gene cluster resides on chromosome 1, differing in localization and ligand specificity. CD1d localizes to late endosomes and lysosomes via a tyrosine-based motif in its cytoplasmic tail.
