• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature (RT).
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at RT for 20 min.

Target Information

STAT3 (Signal Transducer and Activator of Transcription 3) belongs to the STAT protein family, which are phosphorylated by receptor-associated kinases and translocate to the nucleus to act as transcription factors in response to cytokines and growth factors.
Coactivators such as CREB-binding protein are required for transcriptional activation by STAT3.
STAT3 can be activated by Interferon-alpha, Interferon-gamma, EGF, PDGF, and IL6.
Phosphorylation on tyrosine 705 by JAK1 and JAK2 is essential for dimer formation, nuclear translocation, and DNA binding activity.
In humans, the STAT3 gene is located on chromosome 17q.
STAT3 mediates the expression of various genes involved in cell growth and apoptosis.
It interacts with c-Jun, CREB, Rac1, and PIAS3, and has three alternatively spliced transcript variants.
Deletion of the STAT3 gene in knockout mice is lethal at the early embryonic stage.
