• Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, then cool at room temperature for 20 minutes.

Target Information

STAT3 (Signal Transducer and Activator of Transcription 3) is a member of the STAT protein family. It is phosphorylated by receptor-associated kinases, translocates to the nucleus, and acts as a transcription factor in response to cytokines and growth factors. Coactivators such as CREB-binding protein are required for transcriptional activation by STAT3.

STAT3 can be activated by Interferon-alpha, Interferon-gamma, EGF, PDGF, and IL6. Phosphorylation on tyrosine 705 by JAK1 and JAK2 is essential for dimer formation, nuclear translocation, and DNA binding. The human STAT3 gene is located on chromosome 17q. STAT3 is involved in regulating genes responsible for cell growth and apoptosis, and its deletion in mice is embryonically lethal.
