• Antibody is tested in direct ELISA.
• For Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

Cell-mediated immune responses are initiated by T lymphocytes stimulated by peptides bound to MHC molecules on antigen-presenting cells (APC).
Activated T cells express receptors such as PD-1 (also known as PDCD-1) that mediate inhibitory signals from APC.
PD-1 binds two related ligands, PDL-1 and PDL-2, and upon binding, inhibits immune activation in the absence of danger signals (e.g., LPS).
PD1-deficient mice exhibit hyperactivated immune systems and autoimmune diseases.
Although PD-1 has a predicted molecular weight, it often migrates at a higher molecular weight in SDS-PAGE.
