Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- For formalin-fixed tissues: Heat sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, then cool at room temperature for 20 minutes.
Target Information
T-cell–mediated immune responses are initiated by T lymphocytes stimulated by peptides bound to MHC molecules on antigen-presenting cells (APC). Activated T cells express PD-1 (PDCD-1), which delivers inhibitory signals upon binding to its ligands PDL-1 or PDL-2. These interactions regulate immune activation, preventing excessive responses. PD1-deficient mice show hyperactivated immune systems and autoimmune phenotypes. PD-1 often migrates at a higher molecular weight in SDS-PAGE than predicted.
