Target Information

TNFR1 (Tumor necrosis factor receptor 1) belongs to the tumor necrosis factor superfamily and is one of the primary TNF-alpha receptors. It mediates cytokine signaling, positive regulation of the NF-κB pathway, angiogenesis, and negative regulation of gene expression.

The extracellular domain of TNFR1 can be released into circulation as soluble TNFR1 (sTNFR1). In humans, the TNFR1 gene is located on chromosome 12. TNFR1 interacts with proteins such as BCL2-associated athanogene 4 (BAG4/SODD), TRADD, and TRAF2, which regulate receptor-mediated signal transduction.

Mutations in the extracellular domain of TNFR1 are associated with autosomal dominant periodic fever syndrome, likely due to impaired receptor clearance.
