Target Information

ERN1 serine/threonine kinase, also known as endoplasmic reticulum to nucleus signaling 1 (IRE1), is a human homologue of the yeast Ire1 gene product. It possesses intrinsic kinase and endoribonuclease activities and plays a key role in ER-based stress signaling. ERN1 regulates IRE1 proteolysis in mammalian cells and is essential for fetal viability due to its function in extraembryonic cells.
IRE1 acts as a sensor of unfolded proteins in the ER, transmitting stress signals to the nucleus by splicing XBP1 mRNA into a potent transcriptional activator of the unfolded protein response. Null mutants remain viable under normal conditions but are nonviable under ER stress. Expression of a dominant negative form of IRE1p leads to apoptosis when unfolded proteins accumulate in the ER.
