Target Information
ERN1 serine/threonine kinase, also known as endoplasmic reticulum to nucleus signaling 1 (IRE1), is a human homologue of the yeast Ire1 gene product. It possesses intrinsic kinase and endoribonuclease activities and plays a key role in ER-based stress signaling. ERN1 regulates IRE1 proteolysis in mammalian cells and is critical for extraembryonic cell function essential for fetal viability.
IRE1 acts as a sensor of unfolded proteins in the ER, initiating the unfolded protein response (UPR) by splicing XBP1 mRNA into a potent transcriptional activator. Null mutants are viable but essential under ER stress conditions. Depletion of IRE1p does not affect transfected cells under normal conditions but induces apoptosis under stress that causes unfolded protein accumulation.
