Target Information
ERN1 serine/threonine kinase, also known as endoplasmic reticulum to nucleus signaling 1 (IRE1A), is a human homologue of the yeast Ire1 gene product. It possesses intrinsic kinase and endoribonuclease activities and plays a key role in endoplasmic reticulum (ER)-based stress signaling.
ERN1 regulates IRE1 proteolysis in mammalian cells and is critical for fetal viability through its function in extraembryonic cells. IRE1 acts as a sensor of unfolded proteins in the ER, initiating the unfolded protein response by splicing XBP1 mRNA into a potent transcriptional activator.
While null mutants are viable, IRE1 is essential under stress conditions that cause unfolded protein accumulation in the ER. Depletion of IRE1p via a dominant negative form has no effect under normal conditions but induces apoptosis under ER stress.
